Inhibitory Effect of Transforming Growth Factor-¥â1 on Cellular Proliferation and c-myc Gene Expression in a Chriocarcinoma Cell Line (BeWo)


Abstract
Transforming growth factor¥â1(TGF-¥â1) is a potent growth inhibitor for many cell types, including most epithelial cells. We studied the effect of TGF-¥â1 on choriocancinoma cell line in vitro. The effects of TGF-¥â1 were determned on cell
proliforation
and c-myc mRNA expressio by BeWo cell line. TGF-¥â1 inhibited the growth of BeWo cells in a dose dependent manner (P=0.02) as measured by MTT ( [3-4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide]) assay. When BeWo cells were exposed to
10
ng/ml of TGF-¥â1, c-myc oncogene expression was suppressed after 4 hr and completely abolished at 24 hr of stimulation by Northern blot analysis. These results show that TGF-¥â1 may play an antiproliferative effect on BeWo cell growth by
rfeducing
c-myc
gene product. In vitro analysis of cellular and molecular alterations in BeWo cells in this study may aid in elucidating the mechanism of action of TGF-¥â1 on human choriocarcinoma tissues in vivo.
